【摘要】:Theantimicrobialactivityofasubstrate-boundantimicrobialisdependentupondirectcontactofmicrobeswiththeactivechemicalagent.Thistestdeterminestheantimicrobialactivityoftreatedspecimenbyshakingsamplesofsurfaceboundmaterialsinaconcentratedbacterialsuspensionforaonehourcontacttimeorothercontacttimesasspecifiedbytheinvestigator.Thesuspensionisseriallydilutedboth.beforeandaftercontactandcultured.Thenumberofviableorganismsinthesuspensionisdeterminedandthepercentreductioniscalculatedbasedoninitialcountsoronretrievalsfromappropriateuntreatedcontrols.Note28212;Thismethodisintendedforthosesurfaceshavingapercentreductionactivityof50%to100%forthespecifiedcontacttime.1.1Thistestmethodisdesignedtoevaluatetheresistanceofnon-leachingantimicrobialtreatedspecimenstothegrowthofmicrobesunderdynamiccontactconditions.Thisdynamicshakeflasktestwasdevelopedforroutinequalitycontrolandscreeningtestsinordertoovercomedifficultiesinusingclassicalantimicrobialtestmethodstoevaluatesubstrate-boundantimicrobials.Thesedifficultiesincludeensuringcontactofinoculumtotreatedsurface(asinAATCC100),flexibilityofretrievalatdifferentcontacttimes,useofinappropriatelyappliedstaticconditions(asinAATCC147),sensitivity,andreproducibility.Thistestalsoallowsfortheversatilityoftestingcontaminationduetosuchthingsashardwater,proteins,blood,serum,variouschemicals,andothercontaminatesorphysical/chemicalstressesormanipulationsofthespecimensofinterest.1.2Surfaceantimicrobialactivityisdeterminedbycomparingresultsfromthetestsampletosimultaneouslyruncontrols.1.3Thepresenceofaleachingantimicrobialisbothpre-andpost-determinedbythepresenceofazoneofinhibition.1.4Thistestmethodshouldbeperformedonlybythosetrainedinmicrobiologicaltechniques.1.5Thisstandardmayinvolvehazardousmaterials,operations,andequipment.Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
